چکیده
objective: methicillin-resistant staphylococcus aureus is a major cause of nosocomial and community-acquired infections. aminoglycosides are potent bactericidal agents that are often used in combination with either a β-lactam or a glycopeptide, especially in the treatment of staphylococcal endocarditis. the main mechanism of aminoglycoside resistance in staphylococci is drug inactivation by cellular aminoglycoside-modifying enzymes. the main aim of the present study is determining the prevalence of ant(4′)-ia gene encoding one of the most important aminoglycoside-modifying enzymes and simultaneous detection of meca gene responsible for methicillin resistance in clinical isolates of staphylococcus aureus by multiplex-pcr method. materials and methods: a total of 100 clinical s. aureus isolates were collected from shariati and baqiatollah hospitals in tehran, then antibiotic susceptibility pattern of strains were determined by disk diffusion method using penicillin, oxacillin, vancomycin, tetracycline, erythromycin, gentamicin, tobramycin, amikacin, netilmicin and kanamycin disks, considering clsi principles. using agar dilution method the mic for oxacillin, gentamicin, tobramycin and amikacin were also determined. in order to detect resistance genes, ant(4′)-ia and meca, two pairs of specific primers were used and their prevalence was determined by using a multiplex-pcr method. results: all strains were resistant to penicillin (100%) and after that the highest rate of resistance was observed against kanamycin (68%), tetracycline (61%), erythromycin (56%), tobramycin (53%), gentamicin (52%), amikacin and oxacillin (48%) and netilmicin (22%), respectively. all of the strains were also susceptible to vancomycin. in agar dilution method 50% of strains were oxacillin resistant and 49%, 45% and 51% of the strains showed resistance toward gentamicin, amikacin and tobramycin, respectively. thirty-seven percent of the strains also showed high-level gentamicin resistance with mic of ≥128µg/ml. in multiplex-pcr method 53% of the strains possessed meca gene and 58% of the strains were ant(4′)-ia positive. conclusion: results obtained by phenotypic and genotypic antibiotic susceptibility determination tests show that there is a statistically meaningful relationship between methicillin resistance and aminoglycoside resistance in mrsa strains (p

Detection of Methicillin-Resistant Staphylococcus Aureus by Phenotypical and Molecular Methods among Clinical Isolates
Abstract Background and Objective: Increasing prevalence of methicillin resistant Staphylococcus aureus strains (MRSA) with their multidrug resistance potential causes difficulties in the treatment of infections due to these bacteria. Hence, the detection and determination of the frequency of MRSA strains via phenotypical and molecular methods is necessary in different parts of the county. ...
متن کاملPrevalence of erm Gene among Clinical Isolates of Staphylococcus aureus in Shahrekord, Iran
Background: Development of drug resistance to Staphylococcus aureus (S. aureus) has led to the use of older antibiotics such as macrolide-lincosamide-streptogramin B (MLSB) for the treatment of infections. MLSB resistance can be caused by several mechanisms, however, one of the predominant reasons is target modification mediated by erm gene...
متن کاملPrevalence of Vancomycin Resistance in Methicillin-Resistant Staphylococcus Aureus
Background & Aims: Staphylococcus aureus is one of the most common pathogens in nosocomial infections. Vancomycin is the most important therapeutic drug of choice for treatment of infections caused by methicillin-resistant Staphylococcus aureus (MRSA) strains. Therefore, vancomycin-resistant Staphylococcus aureus (VRSA) or vancomycin-intermediate Staphylococcus aureus (VISA) strains are warning...
متن کاملGenetic diversity of Staphylocoagulase genes (Coa) among methicillin-resistant Staphylococcus aureus isolates at clinical specimens of blood and urinary infections
Background: Staphylococcus aureus is a common pathogen in human that can be the cause of a wide range of infectious diseases including bacteremia, pneumonia, cellulitis, and osteomyelitis and skin and soft tissue infections. The coagulase enzyme is one of the most important virulence factors of this bacterium. The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) Coa...
متن کاملDetection of icaABCD Genes in Clinical Isolates of Methicillin-Resistant Staphylococcus aureus from Patients in Iran
Introduction: Methicillin-resistant Staphylococcus aureus (MRSA) is an important pathogen that causes several nosocomial or community-acquired infections. Adhesion to surfaces and subsequent biofilm formation are the major phases of a staphylococcal infection. The aim of this study was to detect the presence of icaABCD genes in clinical isolates of MRSA. Methods: A total of 110 clinical Staphyl...
متن کاملprevalence of genes encoding bi-component leukocidins among clinical isolates of methicillin–resistant staphylococcus aureus
background: staphylococcus aureus has been recognized as a major human pathogen and is the major cause of nosocomial infections. gamma-toxin, leukocidin and other bi-component toxins are a family of proteins encoded by the hlg and luk-pv, respectively. panton-valentine leukocidin (pvl) is an example of these toxins and causes leukocyte destruction and tissue necrosis. the aim of this study was ...
